Abstract:

OBJECTIVE: To investigate the effect and mechanism of Hongqu (Ultivarietas Oryzae Sativae et Monasci), red yeast rice (RYR) in non-alcoholic fatty liver disease (NAFLD).

METHODS: Mice were fed with high-fat diet for 12 weeks to establish NAFLD models. Low (150 mg/kg) and high (300 mg/kg) doses of RYR and silymarin (positive control, 200 mg/kg) was administered by gavage. Following test of biochemical indexes and liver indexes, hematoxylin and eosin staining was performed. In vitro, normal human liver epithelial cell line cells were treated with oleic acid (OA, 0.2 mM) for 24 h to establish NAFLD cell models, with treatment of RYR (674 µg/mL) and adenosine 5'-monophosphate-activated protein kinase (AMPK) inhibitor Compound C (1 mM). Apoptosis was detected by flow cytometry. Meanwhile, oil red O staining, quantitative real-time polymerase chain reaction, and Western blot methods were conducted.

RESULTS: In NAFLD mice, RYR improved liver pathological damage, reduced total cholesterol (TC), triglyceride (TG), low-density lipoprotein, aspartate transaminase, alanine aminotransferase, liver index, lipid deposition, sterol regulatory element-binding protein 1c (SREBP-1c), fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), and peroxisome proliferators-activated receptor gamma (PPARγ) expression, and up-regulated high-density lipoprotein, PPARα, carnitine palmitoyl transferase 1, p-AMPK/AMPK, and p-ACC/ACC expression. In OA-treated cells, RYR inhibited TC, TG, lipid deposition, apoptosis, SREBP-1c, and FAS expression, and enhanced p-AMPK/AMPK and p-ACC/ACC expression. Moreover, Compound C reversed the above effects of RYR.

CONCLUSION: RYR may improve NAFLD by modulating lipid metabolism through AMPK/SREBP-1c pathway, providing a theoretical basis for its clinical application in NAFLD treatment.
