Abstract:

OBJECTIVE: To investigate the role of Longgu (Os Draconis) in the physicochemical compatibility of Chaihu Longgu Muli decoction (chaihulonggudulitang).

METHODS: Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), high-performance liquid chromatography (HPLC), and ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) were used to determine particle size, polydispersity index, zeta potential, pH, viscosity, surface tension, and chemical components of the Chaihu Longgu Muli decoction and comparator decoctions.

RESULTS: It was discovered that Chaihu Longgu Muli decoction forms a smaller, more uniform, and stable particle structure. By comparing measurements of pH values, liquid viscosity coefficients, surface tension, and the yield of freeze-dried powder, it was concluded that this structure promotes the dissolution of components, indicating a more complex interaction than a simple physical mixture Using techniques such as FTIR, ultraviolet spectroscopy, HPLC, and UPLC-Q-TOF-MS, it was demonstrated that Longgu (Os Draconis) can synergistically interact with components such as triterpenoid saponins, phenolic acids, and flavonoids in herbal medicines through hydroxyl and carboxyl groups. These interactions do not affect the types of components in the system but contribute to the stability and dissolution of the components of Chaihu Longgu Muli decoction.

CONCLUSION: This research preliminarily reveals the role of Longgu (Os Draconis) in compound compatibility, offering insights for the modernization of Traditional Chinese Medicine and contributing to the theoretical framework of compound compatibility.
